After reaching the correct pH, I added the agar (gelling agent) at 7 g/L, then placed the media in the autoclave.  Sterilizing all tools, media, and beakers are essential for successful micropropagation.  This step takes about 25 minutes for a liquid cycle.  The machine heats and pressurizes its contents at 250F degrees and 15 pounds per square inch, killing any bacteria or fungus will disrupt the growth of explants.
